چکیده

Abstract The formation of amyloid fibrils various amyloidogenic proteins is dramatically enhanced by ultrasound irradiation. To apply this phenomenon to the study protein aggregation science and diagnosis neurodegenerative diseases, a multichannel irradiation system with individually adjustable conditions necessary. Here, we develop sonochemical reaction system, where an ultrasonic transducer placed in each well 96-well microplate perform sample solutions under high reproducibility, applied it fibril ? , ? -synuclein, 2-microglobulin, lysozyme. results clearly show that our instrument superior conventional shaking method terms degree acceleration reproducibility reaction. controllable controlling driving voltage transducer. We have thus succeeded developing useful tool for proteins.

Mechanisms of amyloid fibril formation by proteins

Understanding the structural heterogeneity inherent in the process of amyloid fibril formation is an important goal of protein aggregation studies. Structural heterogeneity in amyloid fibrils formed by a protein manifests itself in fibrils varying in internal structure and external appearance, and may originate from molecular level variations in the internal structure of the cross-β motif. Amyl...

Amyloid fibril formation by an SH3 domain.

The SH3 domain is a well characterized small protein module with a simple fold found in many proteins. At acid pH, the SH3 domain (PI3-SH3) of the p85alpha subunit of bovine phosphatidylinositol 3-kinase slowly forms a gel that consists of typical amyloid fibrils as assessed by electron microscopy, a Congo red binding assay, and x-ray fiber diffraction. The soluble form of PI3-SH3 at acid pH (t...

Surface-catalyzed amyloid fibril formation.

Light chain (or AL) amyloidosis is characterized by the pathological deposition of insoluble fibrils of immunoglobulin light chain fragments in various tissues, walls of blood vessels, and basement membranes. In the present investigation, the in vitro assembly of a recombinant amyloidogenic light chain variable domain, SMA, on various surfaces was monitored using atomic force microscopy. SMA fo...

Effect of Surfaces on Amyloid Fibril Formation

Using atomic force microscopy (AFM) we investigated the interaction of amyloid beta (Aβ) peptide with chemically modified surfaces in order to better understand the mechanism of amyloid toxicity, which involves interaction of amyloid with cell membrane surfaces. We compared the structure and density of Aβ fibrils on positively and negatively charged as well as hydrophobic chemically-modified su...

Pathways and intermediates of amyloid fibril formation.

The lack of understanding of amyloid fibril formation at the molecular level is a major obstacle in devising strategies to interfere with the pathologies linked to peptide or protein aggregation. In particular, little is known on the role of intermediates and fibril elongation pathways as well as their dependence on the intrinsic tendency of a polypeptide chain to self-assembly by beta-sheet fo...
